Here are five possible interview questions that could be asked in the technical round for the Junior Research Fellow position in Plant Genomics:
- Can you briefly explain the objective and scope of the SERB-sponsored research project you will be working on? Answer: The objective of the project is to investigate the influence of Δ6Des and Δ6Elo genes on Omega 3/6 fatty acid biosynthesis in groundnut using a genetic engineering approach. The scope involves studying the genetic manipulation of these genes and analyzing the impact on fatty acid biosynthesis.
- How would you design an experiment to assess the expression levels of Δ6Des and Δ6Elo genes in groundnut plants? Answer: One possible approach would be to isolate RNA from groundnut plant tissues, perform reverse transcription to obtain cDNA, and then use quantitative PCR (qPCR) to measure the expression levels of Δ6Des and Δ6Elo genes. Primers specific to these genes can be designed and used for qPCR analysis.
- What techniques and methods would you employ to analyze the fatty acid profiles in groundnut plants after genetic manipulation? Answer: Gas chromatography (GC) can be utilized to analyze the fatty acid composition. The groundnut samples can be processed to extract fatty acids, followed by derivatization and analysis using GC. The obtained chromatograms can then be compared to determine any changes in fatty acid profiles resulting from the genetic manipulation.
- How would you ensure the stability and viability of genetically transformed groundnut plants during long-term experiments? Answer: To maintain the stability and viability of genetically transformed plants, it is important to use appropriate selection markers to identify and propagate the transformed plants. Regular monitoring of the plants, proper media composition, and optimized culture conditions are essential for long-term experiments. Additionally, maintaining a sterile and controlled environment can help minimize contamination and ensure the health of the plants.
- Can you discuss a previous research experience or project related to plant biotechnology and its outcomes? Answer: The candidate should highlight any previous research experience they have in the field of plant biotechnology. They can discuss a specific project, the techniques they employed, the objectives, and the outcomes or findings of the research. This will demonstrate their practical knowledge and ability to conduct research in the relevant area.
It’s important to note that the answers provided are general suggestions and should be tailored based on the candidate’s individual knowledge and experience.
