Project title : Liquid-liquid phase separation of α-Synuclein leading to amyloid fibril formation: Implications in Parkinson’s disease pathology

About the project:

  • Liquid-liquid phase separation (LLPS) is an important phenomenon for explaining the formation of membrane-less organelles in cells.
  • Recent studies also suggested that LLPS is also responsible for amyloid fibril nucleation associated with various neurodegenerative diseases such as Alzheimer’s, Parkinson’s and frontotemporal dementia.
  • This DST project studies alpha-synuclein protein (which aggregates and kills dopaminergic neurons in Parkinson’s disease) phase separation in vitro and inside the neuronal cells.

Here are some potential interview questions and answers for the JRF position at IIT Bombay:

1. Can you briefly explain your educational background and any relevant qualifications or certifications you hold?

Answer: I hold an MSc degree in Biotechnology. I completed my degree with a first-class distinction, which reflects my strong academic background in the field. Additionally, I have hands-on experience in both protein chemistry and cell biology, which aligns well with the requirements of this role.

2. Could you describe your familiarity with Liquid-Liquid Phase Separation (LLPS) and its significance in cell biology and neurodegenerative diseases like Parkinson’s?

Answer: Liquid-liquid phase separation (LLPS) is a fundamental phenomenon in cell biology, responsible for the formation of membrane-less organelles. LLPS has gained significant attention due to its role in the nucleation of amyloid fibrils associated with neurodegenerative diseases, including Parkinson’s. I am familiar with LLPS and understand its importance in the context of this research project. My academic background in biotechnology has provided me with a solid foundation in these concepts.

3. Can you share your experience with protein expression and purification techniques?

Answer: I have practical experience in protein expression and purification techniques gained through both coursework and laboratory work during my MSc program. I have successfully expressed and purified various proteins for different research projects. These experiences have equipped me with the necessary skills to contribute effectively to the protein-related aspects of this project.

4. Could you explain your understanding of the role of α-Synuclein in Parkinson’s disease and how this project aims to investigate it?

Answer: α-Synuclein is a key protein associated with Parkinson’s disease, known for its aggregation and its impact on dopaminergic neurons. This project aims to study the liquid-liquid phase separation of α-Synuclein both in vitro and inside neuronal cells. By doing so, we hope to gain insights into how LLPS contributes to the formation of amyloid fibrils in Parkinson’s disease, ultimately advancing our understanding of the disease pathology.

5. What motivates you to apply for this Junior Research Fellow position at IIT Bombay and how do your qualifications and experiences align with the requirements of the role?

Answer: I am motivated to apply for this position because it offers an excellent opportunity to contribute to cutting-edge research in the field of neurodegenerative diseases, particularly Parkinson’s disease. My educational background in biotechnology, coupled with my experience in protein chemistry and cell biology, aligns perfectly with the job requirements. I am excited about the prospect of working on a DST project that could potentially have a significant impact on our understanding of Parkinson’s disease pathology.

ALL THE BEST!
