Role Description
Sanzyme Biologics is seeking a Biomolecules Downstream Processing Specialist with strong expertise in the purification of recombinant proteins, enzymes, peptides, and biologics (primarily expressed in E. coli). This is a full-time, on-site role based in Hyderabad. The ideal candidate will have hands-on experience in designing, optimizing, and scaling downstream purification processes—from bench scale to commercial manufacturing—under cGMP environments.
Key Responsibilities
Primary Recovery
• Optimize cell harvest operations (centrifugation, microfiltration)
• Execute high-efficiency cell lysis (homogenization, sonication, chemical/enzymatic methods)
• Handle inclusion bodies: isolation, washing, solubilization
Protein Refolding & Solubilization
• Solubilize proteins using chaotropic agents (urea, guanidine HCl)
• Perform high-throughput screening for refolding conditions
• Execute redox-controlled refolding for disulfide-bond-containing proteins
Chromatography Expertise
• Operate and optimize: Affinity (His-tag, GST, Protein A/G, IMAC), Ion exchange (IEX), Hydrophobic interaction (HIC), Size-exclusion (SEC), Mixed-mode chromatography
• Develop multistep purification workflows
Filtration Technologies
• Perform MF and UF/DF operations
• Conduct buffer exchange and concentration
• Scale filtration systems (flat sheet, hollow fiber, TFF)
Process Development & Scale-Up
• Conduct Design of Experiments (DoE)
• Perform process robustness and characterization studies
• Implement viral inactivation (where applicable) and endotoxin control
• Support tech transfer to pilot and commercial manufacturing sites
Quality & Regulatory
• Work within cGMP operations
• Prepare and maintain batch records, SOPs, validation documents
• Conduct purity, potency, impurity profiling
• Contribute to endotoxin reduction strategies and QC method development
Qualifications
• MSc / MTech / BTech in Biotechnology, Biochemistry, or related Life Sciences (Master’s preferred)
• Strong expertise in downstream processing: chromatography, filtration, centrifugation, and precipitation
• Hands-on experience with analytical and QC methods
• Demonstrated ability in process optimization and troubleshooting
• Knowledge of regulatory and compliance standards in manufacturing environments
• Excellent analytical thinking, problem-solving abilities, and attention to detail
• Strong collaboration skills and ability to work in multidisciplinary teams
• Prior experience in probiotics, fermentation, or related biotech fields is a plus
