Answer: In my previous role, I have extensively worked with whole genome sequencing and amplicon sequencing. I have hands-on experience in library preparation, sequencing platforms such as Illumina, data analysis using bioinformatics tools, and interpreting the results to identify genetic variations and mutations.
  2. How have you utilized molecular biology techniques in your research experience? Can you provide examples of specific experiments or projects where you applied these techniques? Answer: During my research experience, I have utilized various molecular biology techniques such as DNA extraction, PCR, cloning, gene expression analysis, and gene editing using CRISPR-Cas9. For example, I have successfully cloned target genes into expression vectors and performed functional studies to analyze gene expression levels and protein interactions.
  3. Tell us about your expertise in flow cytometry and cell sorting. What types of samples have you worked with, and what were the objectives of the experiments? Answer: I have extensive experience in flow cytometry and cell sorting, primarily focusing on characterizing and analyzing cellular populations. I have worked with various cell types, including immune cells and cancer cells, to study their phenotypic markers, cell cycle progression, apoptosis, and intracellular signaling pathways. These experiments aimed to understand cellular dynamics and identify specific cell subsets.
  4. Can you discuss your experience with RNA-sequencing and its applications in your research? How have you handled and analyzed large-scale transcriptomic data? Answer: In my research, I have employed RNA-sequencing to study gene expression profiles and transcriptomic changes in different biological conditions. I have designed and optimized RNA-seq experiments, performed quality control checks, generated transcriptome data, and conducted differential gene expression analysis using bioinformatics tools such as DESeq2 and edgeR. I am familiar with handling large-scale datasets, filtering, normalization, and pathway analysis.
  5. Could you elaborate on your proficiency in mammalian cell culture and confocal microscopy? What types of cell lines or models have you worked with, and what types of imaging experiments have you conducted? Answer: I have extensive experience in mammalian cell culture, including adherent and suspension cell lines. I have performed cell culture maintenance, transfections, drug treatments, and cell-based assays to investigate various cellular processes. Additionally, I have utilized confocal microscopy to visualize subcellular structures, protein localization, and cellular dynamics. For instance, I have studied protein trafficking using fluorescently labeled antibodies and tracked intracellular signaling events using live-cell imaging.
