Primary liver cancer is one of the world’s deadliest cancers. It is the 5th most common cancer worldwide & represents the 3rd most common cause of cancer death. Hepatocellular carcinoma (HCC) accounts for 90% of primary liver cancers & is refractory to nearly all currently available anti-cancer therapies. The incidence of HCC in developed first-world countries has been increasing over the last 20 years; in Australia the incidence for HCC has nearly tripled. The obesity epidemic is thought to account for as much as 50% of the increase in HCC in developed nations. The prevalence & increasing trend of obesity in the developed world will translate into a significant rise in HCC incidence. However, the molecular basis by which obesity promotes HCC is unknown. The current project will take advantage of cell models and conditional knockout mice to determine the impact of obesity on HCC development/progression.
