Experimental Conversions - Important for all Life Science Entrances as well Lab Work
Hi Friends,
We perform lot of experiments in laboratory and find it very confusing at the end to solve the calculation involved and to convert in the desired units it a tedious job. For e.g For a 1 picomolar solution how much amount of solute should be present in the given solvent. or convert microgram per mole to milligram per mole or viceversa. Its really a hectic job to do.So here are a few basic conversions which may be helpful and can make your work easy. In entrances too you will find these conversion units convinient to solve the problems.
Metric Prefixes
Prefix Symbol Factor
kilo k 103
centi c 10-2
milli m 10-3
micro µ 10-6
nano n 10-9
pico p 10-12
femto f 10-15
atto a 10-18
zepto z 10-21
Spectrophotometric Conversions
1 A260 unit of double-stranded DNA=50 µg/ml
1 A260 unit of single-stranded DNA=33 µg/ml
1 A260 unit of single-stranded RNA=40 µg/ml
Protein Molar Conversions
100 pmol of 100 kDa protein = 10 µg
100 pmol of 50 kDa protein = 5 µg
100 pmol of 10 kDa protein = 1 µg
100 pmol of 1 kDa protein = 100 ng
Protein/DNA Conversions
1 kb DNA = 333 amino acids = 37 kDa protein
270 b DNA = 10 kDa protein
810 b DNA = 30 kDa protein
1.35 kb DNA

Hi thank you so much , it is really helpful.
but i have 2 qs
1. how much should be absorbance so that it will correspond to 50ug/ml
2. how to find out average mw of ds DNA base pair?
by measuring spectrophotometricaly / any other method
i know my doubts must be simple for u but i am lacking knowledge about it so i am asking u
if possible plz reply
thanks
absorbance would be 1 Sukhada
absorbance would be 1 Sukhada for 50 ug/ml
Hi
Hi Sukhada,
First of all thanks for appreciating my blog.!! Now as Mr Bachan says koi sawaal chota ya bada nahi hota similarly your query is a valid one..!!
Coming to question no.1
The absorbance should be 1 i.e your instrument should read an O.D. of 1.
Coming to question no.2
No need to do tedious job our scientists have all calculated standard values for us but still if you are intrested in the method of calculation here is is...!!!
DNA is made up of four building blocks, called nucleotides. The molecular weights of
these four nucleotides (A,C,G,T) are not identical, so in routine work we usually
average the molecular weights of all four and go with 330 daltons per nucleotide in
DNA (=330 grams/mole of individual nucleotides). Since DNA is double stranded, i.e.,
one nucleotide pairs with a nucleotide on the opposing strand, then we can say the average molecular weight of a nucleotide PAIR in
DNA is 660 daltons.
Hope I have satisfied your query..!!!
thanks!!!
thanks!!!
The explanation is really
The explanation is really simple and clear. I have to admit..understood this only now. Thanks to Abhilash :-))
